Advanced chemistry, optics, and informatics combine to deliver exceptional speed and data quality, outstanding throughput and scalability.
Optimized reagent kit that provides increased cluster density and read length, improving sequencing quality scores compared to earlier versions.
MiSeq Reagent Kits v3 enable the highest output of all MiSeq kits. The v3 kits retain the same pre-filled, ready-to-use reagent cartridges as the v1 and v2 kits, but offer improvements to enable increased cluster density and read length, as well as higher Q30 scores. The v3 kits offer:
High-quality sequencing and a broader range of sequencing applications on the MiSeq System
Double the amount of output per flow cell and an increase in read lengths
An increase in the number of single and paired-end reads to support a wide range of applications
Radio-frequency identification (RFID)–encoded consumables to ensure reagent traceability and compatibility
Updated system software unlocks the full benefits of the v3 kits. Access the latest software.
This product is also available as an Illumina Advantage (TG) product. Illumina Advantage large-scale sequencing products feature lot-specific shipments and testing, extended shelf life, and advanced change notifications for greater laboratory efficiency.

A MiSeq flow cell is a single lane flow cell that can support up to 25M reads per run.
There are two new reagents in MiSeq Reagent Kit v3, the Incorporation Mix (IMT) and Scan Mix (USM). In addition, the PR2 volume has been increased to 500 ml to support longer runs. This ensures double the output of a single run using the v3 kits (3.3–15 Gb) compared to v2 kits (0.75–8.5 Gb).
The number of samples that can be loaded depends on the expected coverage and genome size.
For assistance, use the Sequencing Coverage Calculator tool.
Yes. As with MiSeq v2 kits, MiSeq v3 kits are compatible with all Illumina library prep kits. Some library prep protocols require slight changes to reach optimum clusters. Illumina recommends a starting concentration of 4 nM to begin the denature and dilute step before loading libraries for sequencing.
Read lengths of 2 × 75 bp can take ~21 hr, while read lengths of 2 × 300 bp can take ~56 hr.
